Linking a GPO

My supervisor and I are in disagreement regarding the linking of a GPO. She has linked a GPO under a OU where there is no other child objects. Will the GPO be processed by the OU it is linked under? For example, there is the Domain then a "Site" OU and within that a "Users" OU which contains several user objects within. The GPO was linked to the Users OU, will this apply to the users in that OU or does it have to be linked above that to the Site OU?

Hopefully this all makes sense, just trying to understand the processing of it all. Thanks for your assistance!
